This is more opinion than theory.
I think John Locke is John Locke but with a purpose. Furthermore, I believe his speech to his people that he would want to see the person giving him orders (Jacob) was another tactic to gain the trust of his followers. Think of the scene in S5E15, when he asked Ben "hasn't the island spoken to you before?" I beleive that he is out to prove that there is no Jacob or @ least the Jacob Ben, Richard speaks of isn't as mighty as the followers think.
So in essence killing Jacob isnt a physical killing but a spiritual killing (like killing the belief of Jacob). I think that he will expose to his followers that there is no Jacob. That's why Richard and Buggy eyes (I love both characters) is very concerned.
