Here's my thoughts on the Season 6 opening episode:
Flight 815 will crash again, with Jack waking up in the jungle, mirroring the opening of Season 1 (people expect it to be the series' final shot, but I think it'll be mirrored here, not at the end of the season). Everyone has the memories of what happened to them on the island until the bomb went off, but are re-living their experiences...sort of.
When Jack arrives at the beach, he finds that everyone who had died on the island (Boone, Shannon, Ana-Lucia, etc) have now died in the plane crash, course correcting all the deaths and leaving the same main cast as we had in Season 5. Course correction is a bitch.
Except...CLAIRE IS STILL ALIVE, AT THE BEACH-CAMP AND THUS ONE OF THE MAIN CAST AGAIN!!!
Ben can't send Ethan or Goodwin to inspect the crash scenes (since Ethan isn't around due to course correction), so he ventures into the Fuselage camp himself. Jack 'welcomes' him, but reluctantly. Juliet is still at Otherville and comes with Ben.
Things are decidedly awkward between Jack, Sawyer, Juliet and Kate, but I can't be bothered to theorise over this, since the quadrangle drives me nuts.
Only Bernard is at the Tailies section, since the rest died on the island the first time around and in the plane crash the second time around. Jack et al realise this and start a trek to save Bernard, prompted by Rose.
Locke is still around, but as the creepy not-Locke inhabited by whatever force that wanted to take out Jacob. He will become this season's "Big Bad", what with Ben realising he tricked him into killing Jacob and aligning himself with the good guys.
The biggest shock of the opening is that the hatch is still there, and that's the reason they still crashed. What's more, Desmond is still there. He thinks he's just having another consciousness-flash, until Jack convinces him that it's actually happening again.
The season's main storyline is trying to TOTALLY reverse the timeline so as to avoid the crash altogether, as they tried to at the end of Season 5, instead of just catapulting themselves back to the point of the crash. The season will end with the whole of the Jughead bomb being detonated, destroying the island and it's properties entirely, thus actually letting the plane arrive safely.
