Jacob seems to be possibly the most important charecter in the show. He has some unexplained powers and seems to know everyone and everything. After a lot of consideration I have found 3 candidates who could possibly be Jacob.
1. Locke's Father- Locke too seems to be very special and has many unexplained powers. Locke is supposed to be the true leader of the others. Isn't that Jacobs job? I think Locke came to the island to replace Jacob who is getting old and nearing death. Locke was forced to "kill" his father to prove he was ready. Now he is charged with bringing everyone back to the island and to "die" as a final task to show he is ready to rule over the island. Lockes healing abilites were passed down in his family genes. When the losties are reunited he will finally replace his father Jacob and become the islands supreme leader.
2. John/Claire's Father- At the end of season 4 we see Claire and Father mysteriously together in Jacobs Shack. Its quite possible that despite what he said (that he was just communicating for Jacob) that he really is Jacob, hence his "ressurection". He arranged his death so jack would get on the flight and come to the island.
3. Somebody else- its quite possible jaccob is someone else entirely. Theory by PinkFloydPatrick
