First, we know that he is aware of the island in detail. He knew about the Black Rock (log recovered), the plane (flight recorder, a modern equivalent to a ship's log), Ben, etc. And I think we have more than a hunch that he sent Desmond to the island knowingly.
Second, we know he is rich and self-made.
But we do not know HOW he made his money. My theory is that the island repeats itself (which is not a new theory) and that Aaron is the new Mr. Whitmore, who himself is the new Jacob.
Babies conceived on the island can't survive, but babies conceived off the island and born on the island can. Furthermore, we know that Aaron is considered highly special in the future. Therefore, I figure, people born on the island are given a magical quality.
Jacob must have been in utero (inside a pregnant woman sailing on the Black Rock) when the ship arrived. That gave him enourmous powers. Whitmore must have arrived on the island in utero and been born on the island. That gives him powers and knowlege of the island. Unlike Jacob, however, Whitmore choose to return the the mainland and use his powers to build a fortune and attendant power. Now he wants to return the island and kill everyone, to eliminate competition.
This explains why Ben fabricated the story that he was born on the island. It built credibility with Richard et al. He also therefore had to kill everyone associated with Dharma, who knew the truth. This way, Ben established himself as the heir to Jacob. (Jacob was probably originally incapacitated by Whitmore himself, before leaving the island.)
(Could it be that Whitmore himself arrived on the island via a sailboat? Could the sailboat that Desmond tried to use during Season Two be Whitmore's old boat? Is it based on his own experience that he therefore knew that Desmond would end up on the island when he left on his ocean voyage?)
Now, thanks to the Oceanic crash, Aaron has come as a new Messiah for the island. That's why Whitmore has to kill everyone on the island -- a kind of modern-day Harrod trying to kill Jesus. But, as in the Bible, Jesus (Aaron), escapes to return to the mainland to become one of the Oceanic Six.
Whitmore is targeting Ben, who he thinks was born on the island, when in reality he should be targeting Aaron, who actually was!
