We know that the MIB occupied Locke's body somewhere along the way, and that (Fake) Locke convinced Ben to kill Jacob.
We also know that the Others sent bodies out to sea, in order to avoid the bodies being occupied by spirits.
There's an extra Locke body available for the taking the moment that Jacob is killed.
It seems quite possible to me that Jacob will commandeer the extra Locke body and we'll have 2 Locke's floating around, trying to trick each other, and those that are on the island. Eventually they will either face off, or a character (likely Jack) will be forced to make a decision.
It's the classic evil twin setup.
