As the title suggests, I believe Lost is nothing more than a battle between good and evil, a game that Jacob and Esau have been co-ordinating potentially from the beginning of time. Just who is good and who is evil remains to be seen (however i believe that Jacob represents good and Esau represents evil).
The opening of the Season 5 finale reveals exactly this. They 'bring people' to the island (as is said by Jacob when they spot the ship), and constantly test them to find the ultimate victor. Esau says they come, fight, corrupt and destroy. he believes fundamentally that all humans are bad and Jacob is trying to prove him wrong, saying that everything on route to him being proven right (proving there is ultimately goodness), is just progress. Esau has had enough of the game (for want of a better term), and wants to kill Jacob to end it all.
The island is a paradise (a garden of eden represented by the adam and eve corpses discovered in season 1) and the ultimate prize. Everyone who comes to the island - or has been there in the past - is drawn into this game, of which there is yet to be a winner. However at the end of season 6, there is to be a conclusion to the game, with it's current players ultimately deciding the outcome. Jacob and Esau are always represented by two people who are appointed leaders of both sides. Throughout Lost there has always been a protagonist and an antagonist struggling to overcome and claim victory over the other. Jacob and Esau, Ben and Widmore, and Jack and Locke being the final two representatives.
neither one can kill the other. As is demonstrated by Esau admitting to needing to find a loophole to get Jacob. In series 4, ep.9 - The Shape of Things to Come - Widmore asks Ben if he is going to kill him, and Ben replies that they both know he cannot do that. In season 6, Jack and Locke will clash I am certain, and we will discover they cannot kill each other either. It is what the 'leaders' represent and the people who they have on their side, who will ultimately decide the victor in the eternal struggle.
In series 1, ep.13 - Hearts and Minds - Jack and Locke have an amazingly similar conversation to Jacob and Esau where Jack sits down by the sea next to Locke, and begins a conversation that starts with 'are there any ships?' This blatant comparison with the opening of season 5 cannot be a coincidence, and reinforces my theory, and is the earliest indicator of representation of both sides. It is interesting to note the reversal of who I believe represents good and evil. Locke is already sitting down like Jacob was, and Esau walks in like Jack does. Perhaps this is the twist that we can expect in Season 6. Locke is actually the good guy and Jack the bad guy.
