I spent a few minutes putting this together only to discover others including Chim Richalds has posted something similar but I will post nonetheless. Like many other John Locke is my favourite character in the show. I root for him every time he is on screen and I did hope that he would find his purpose in life.
However I now come to the conclusion after watching Dead is Dead and Follow The Leader that the John Locke who crashed on the Island is Dead. I believe that he died in the hotel room when Ben strangled him. The resurrected shell of John Locke has been inhabited by something else. My deduction leads me to believe that what’s inside John is a malevolent force perhaps the Smoke Monster. What cemented this belief for me is not that he wants to "kill" Jacob but that cruel manner he has treated Sun pretending to care for her and her cause but just stringing her along to follow his own dark agenda. Perhaps she maybe the cast member who dies as he uses her as tool in attempted killing of Jacob.
I feel Jacob who once led The Others is now a prisoner perhaps imprisoned by Ben. The Others have taken a wrong course under Ben and I feel they now represent the Dark Side to borrow a phrase from Star Wars. There is a war coming and I feel this war will be at the heart of Season 6. John Locke and The Others are on the wrong side they have been misled by darkness. If John succeeds in killing Jacob this will bring earth to the brink of destruction. The story of Lost seems to be in time loop with some people trying to prevent the destruction of earth while others welcome it. What is in John seeks to follow a destructive path. Jack actions will cause The Incident and for the Oceanic losties to move from 1977 to the future. They will be on one side of the war and John Locke and his band will be on another. The reason why Richard is ageless is because he is Time Guardian and serves a higher force.
The John Locke I rooted for is now more. He lived ultimately an unfilled life to use his own words "pathetic one" and now an evil force inhabits his shell. Perhaps where he went wrong was that he didn't turn the wheel the first time. Perhaps Ben knew if John turned the well things may have taken another direction.
