Jacob wants Locke to kill him. Jacob currently doesn't seem to "exist" in the physical sense. We have seen a brief moment of him, we have heard him, we have seen others speak his name, but never seen him. Christian spoke "on his behalf" because Jacob is unable to. John Locke died and he was resurrected. If Locke kills Jacob, in whatever way he can or knows how, the island will resurrect Jacob and he will be in the physical form again, maybe in a John Locke sense or maybe how Christian appears.
Either way, if Jacob is all knowing all seeing all powerful, then he must know of Locke's plan or at least be more powerful to stop him. He could easily take his "life" away or his ability to walk. The only conclusion is that he wants it to happen. "Help Me" he said to Locke in Season 3. This is helping him. He have not seen any advancement on that part of the story.
