I believe the Incident will be a large discharge of radiation that will affect anyone nearby in the same way the hatch implosion affected Desmond. These affected characters will then have the ability to travel in time via their consciousness, as we've seen Desmond do. I believe this method of consciousness TT is "naturally occurring" and uncontrolled.
I also believe the Others over time have been able to practice a controlled version of the phenomenon whereby they can willfully transport their consciousness to particular points in time and perhaps even to particular people.
I support this theory by citing two unsolved mysteries in addition to the Incident: What is the Temple, and What are the Whispers?
These mysteries seem to have been introduced to set up some big reveals. As such, I think they both play a big role.
If this theory is correct, then the Incident revelation may be of the kind where you are compelled to rewatch past episodes -- in this case to witness characters experiencing consciousness TT and to examine what "rules" may apply.
Perhaps loss/gain of consciousness is a factor. Many characters have experienced strange things upon being knocked out, sleeping/dreaming, drinking, undergoing anesthesia. For example, Hurley wins the lotto, loses consciousness, then says he doesn't want anything to change, as if he knows the future. Jack does not want to be put under during his appendectomy. Claire's nightmares and visions.
Perhaps characters look or behave differently when "occupied" by another consciousness. For example, other theories have noticed changes in eye color at times. Characters comment on how someone seems different or new, as when Kate says she doesn't like the new Jack, or when Charles tells Ben he "knows what you are, boy," or when Horace fire-bombs the saplings.
Now the Temple revelation. As other theories have stated, this may hinge on contrast: behind the ancient walls is hidden something highly technical. Perhaps a technology that allows the Others to control this type of TT. Perhaps technologies that also control other aspects of what we've seen on the Island (Smoke Monster? Reanimation? Physical TT?). DHARMA may have been studying "natural" properties of the Island that were actually being harnessed/generated by the Others.
Finally, the Whispers revelation may be one of recognition (if and only if you've read the Whispers transcripts). Correct me if I'm wrong, but we have not heard a single line of the Whispers spoken by a character in normal dialog. As soon as we who have read the transcripts hear one of those lines (like "How could you say that? I knew he was American") we'll instantly know the source of the Whispers in a collective "a-ha" moment.

- There are authorized and non-authorized whisperers
- Whisperers are afraid of being seen by people
- Whisperers are afraid of being caught by people
- Whisperers are afraid of being caught by other whisperers
- They reference some sort of building with technology
- Our Losties are also whisperers
- They try to recognize familiar and friendly people
- They have trouble maneuvering in real space
- They can control whether people see and hear them
- They need help
- They are carrying out a mission ... to "get" something out of people?
- There are experienced whisperers (in control) and novice whisperers learning the procedure
- They can access people's minds
(I plan to gather excerpts supporting each observation in a separate theory.)
If my theory is right, then I would expect to see Season 5 end with the Incident revealing one example of consciousness TT (perhaps Jack in the pilot?) and Season 6 end with a full reveal and explanation of the Temple technology and its operators.
