I don't expect that we will find out who Jacob is until next season, possibly in the final episode, but I thought I'd run down the candidates, presuming that he's someone we know. A lot of this assumes that the Losties go back in time to Jacob’s origins, which may or may not happen. Still, it’s an interesting exercise.
Locke – Locke’s case is that he already has the strongest connection to the Island, and if he’s meant to be leader of the Others, he may have been destined to be the ultimate leader, Jacob. He’s also special, not because he believes that he believes in his destiny, but because Jacob (himself?) called out to him for help. It may be that Locke’s body, through coming back to the Island, becomes reincarnated as Jacob.
Jack – Possibly the Jacob to Locke’s Esau. Esau was a hunter, while Jacob was more of an indoor guy. Locke, like Esau, joins the Others (the Canaanites), though not through marriage. Jacob flees from his home, but eventually returns and becomes the founder of the Israelites. If the Others are destined to go back in time to become the original Others, this might be Jack’s role in that scenario (or he’s Moses).
Aaron – Aaron’s birth plays a major part in the narrative, which gives him a level of importance that very few of the other characters have (the others being Locke, Ben, Ji Yeon, and Desmond’s Charlie). His birth on the Island was possibly the first in 16 years. He seems to be a key element in the struggle between the two forces on the Island, one bringing him there and the other arranging for him to be separated from his mother and leaving the Island.
Sawyer – Sawyer has had the most redemptive arc on the show, and sacrificed himself for others. He also killed his symbolic father, an act that is held in some esteem by the Others. Plus, his name is James, and James is derived from the name Jacob.
Christian – Probably not, as he’s already denied being Jacob, just speaking for him. Actually, I’m not even sure he does that.
Jin – Along with all the other male 815ers, he can’t be completely discounted, though I think it’s unlikely. His story is too tied up with Sun’s.
Desmond – He can’t be completely discounted yet, though I think he’s more likely destined to be Adam. Still, he is special, and the rules of space and time don’t apply to him, which would seem to fit Jacob’s profile. I see his fate more tied up with Penny’s, though.
Walt – Special, like Desmond and Locke, though he seems more like a profit than Ghost Leader.
Widmore – If Jacob turns out to be evil, he’d be a good candidate. Still, very unlikely.
