There has been a lot of speculation as to the identity of Jacob. There are countless theories posted on this web site alone. But there is a video posted on YouTube that helps to confirm my theory of who Jacob really is. I'll get to that in a minute.
I'm not sure where or when this bit of info was revealed by The Powers That Be, but I read somewhere that the role of Jacob has not been cast yet. But I don't think that necessarily means the appearance of Jacob will not be a familiar one. I think it just means that Jacob has not been seen before specifically on the television program called "Lost."
There is a video posted on YouTube that uses video manipulation techniques to tweak the scene in Jacob's Cabin where we get a glimpse of his image. The video uses this technique to "prove" that the image of Jacob reveals him to be John Locke. But I don't think so. I believe Jacob is someone not before seen on the television show, but is a large part of the Lost universe, specifically from The Lost Experience. I believe Jacob is actually Dr. Thomas Werner Mittelwerk.
Those of you who have gone through the "entire" Lost experience will recognize that name both from Dharma films seen on the show, and from the alternate reality game The Lost Experience. For more background on Mittelwerk, just go to Lostpedia.
I believe that Mittelwerk, i.e. Jacob, was one of the original inhabitants of the island. He may or may not have been on the Black Rock when it was thrown onto the island in the 1800s, if you believe that they were the original inhabitants of the island. He is one of the Hostiles, and, like Richard Alpert, he does not age. He has positioned himself into the Dharma Initiative supposedly to work on solving the Valenzetti Equation. However, I believe he is actually working to PREVENT the equation from being solved. Because I believe "Jacob" will live either forever, or until the Valenzetti Equation is solved, whichever comes first. So, as long as Jacob is kept alive, the world will not come to an end, basically.
I haven't worked out the fine details yet, because I don't think I have enough information to prove it, and I probably won't until at least the end of season 5. But when I saw the image of Jacob go through its transformation in that YouTube video, I immediately saw Dr. Thomas Mittelwerk.
