Jacob said 'Help Me' to Locke because he is suspended in time and can only interact through others, the smoke monster etc.
He wants to die, he wan't to go. Jacob is great but the island is greater and the island won't let him die until his job is done. Jacobs job is to protect the island and find a replacement... Locke! He's found Locke over a long process of recuiting people to the island, people who will believe in the island and work for him.
These people, lead by Ben on island and Richard off island (most of the time) used time travel to find the possible candidates to replace Jacob. They started by bringing one person at a time and carrying out socail experiments i.e. Desmond in the hatch.
At the same time Jacob had to keep the island safe, hidden and pritected by the his team of the others. Ben was a good leader and when he got the tumour, Jacob had to find the best spinal surgeon around... Jack. Wihtout Ben as a leader, the others may fault to protect the island.
The others would go back in time to recruit both people to work on and protect the island as well as possible candiates to replace Jacob. They would control a series of events in the past, even from birth, all of which would lead them to the Island. All 815's would have no reason to go back due to these events and therefore would be willing to stay, and eventually become the replacements for the others or added to the team. Amony these people would be a great leader... jack & Locke, at the time either could have replaced Jacob but Jack, no matter how teseted he was just couldn't take the lead of faith and believe in the island. Locke, altough o-yoing with faith, eventually became one with the island and was ready to replace Jacob, within time.
Ben was only ever there to lead the others and eventually to find Jacob's replacement. After this he could leave, altought he really didn't want to as he'd spent most of his life there. Ben, always having a plan, knew that he could be allowed to come back if he had work to do. That's why he let some of the 815's go, as hew knew oneone should leave the island until their job is complete. Know in season 5 Ben will continue to manipulate people while working with Locke who is alive on the island and eventually get everyone to come back. Locke sacrifysed him self by leaving the isand to save the island. He then went to the O6 individually and told them all that they must return, or told them what they needed to know to make them meet up and return. He then killed himself in the hope that he would be reivived by the island upon return in the coffin. then onto season 6
