Possible spoilers in this theory.
It's obvious that Locke is one of, if not the most important characters on Lost. He also has one of the closest connections to the Island and since S1 he's been trying to figure out the answers of the Island. Now let me ask you this, if your legs couldn't function and you were told that you would be in a wheelchair for the rest of your life, then miraculously survive a plane crash and landed on an island, then suddenly you can walk again...would you be completely changed as a person and have faith in the island? Well Locke has certainly changed by looking at his flashbacks and he's also growing on the island as well. In S1 he's trying to find the meaning of why he was brought to the island, in S2 he struggles with his faith in the island, S3 he becomes more closer with the island, and in S4 he has taken on a great deal of leadership.
Ever since he was healed by the island he has mostly kept it a secret and has been trying to unravel the mysteries of the island and what his purpose is there. He has done several great acts for the castaways there. He's helped Charlie overcome his drug addiction which probably saved him, he provided food for the camp by hunting boar, used his tracking skills to trek through the jungle and search for missing people, help Boone with his problems with Shannon, built a cradle for Aaron, befriended several of the castaways, and many other things as well.
Since S1, he has been looked at as a villian and not trustworthy, mostly through Jack's perspective ever since Boone's death. Locke and Jack pushed the button in S2 which the two feuded a great deal during that time. In S3 when Locke proposed to go and rescue Jack after been having held prisoner of The Others. On the mission, he also looses trust from Sayid and Kate as he lies to them. When they get to Otherville, Locke then blows up the submarine which is their only way of the Island, so that nobody can escape. This made Jack furious. It has never been revealed so far as to why Locke did that, but I'll get to why he did that later.
He then leaves Otherville with The Others, but never makes an alliance with them. He's there to observe and get answers. Skip ahead to later and eventually Ben shoots Locke and leaves him for dead. Later on, Locke woke up in the grave he fell in and is unable to move his legs. Because he can't move and is in a great deal of pain, he reaches for a gun and is about to kill himself. Then suddenly Walt appears and tells him to stop and that he has work to do. While all of this is going on and Locke was away from the castaways at the beach, they encountered Naomi who said that she was part of a rescue team and was soon going to rescue them.
At the radio tower, just as Naomi is making the call to save them Locke throws a knife in her back, then points a gun at Jack to not answer the phone or he'll kill him. Jack calls Locke's bluff and answers the call, then Locke takes off into the jungle. Now through the castaways perspective, Locke is a murderer and villian. If Locke was really a villian, why didn't he just kill Jack?
Locke is actually there to save them. Not only is he trying to save them by preventing them from leaving, he's also trying to protect them from the freighter crew. Eventually the castaways split into two camps , Team Locke and Team Jack. Locke believes that the freighter crew coming to the island are actually the bad guys, and he doesn't want to be rescued. Jack thinks he's crazy and that they are all going to be okay.
So exactly why is Locke preventing them from leaving? He wants to protect them and save them all. Here's some evidence:
1) In the flashforwards when Jack is off the island, he is actually worse off than he was before. He's an alcoholic, drug abuser, doesn't really care about anybody or saving lives, and suicidal. He desperately wants to go back to the island.
2) In Hurley's flashforwards when he's off the island, he eventually ends up in a mental institution and he also wants to go back to the island.
So you can see that Locke is trying to be a hero and make them stay on the island. So is the freighter crew there to save them or kill them? Well as we know in this weeks episode, they attack Team Locke's camp trying to kill about everybody. So I guess Locke is right, huh Jack?
